Pfu DNA Polymerase is a high-fidelity, thermostable DNA polymerase originally isolated from Pyrococcus furiosus, a hyperthermophilic archaeon, and expressed recombinantly in E. coli. Its defining feature is a highly active 3' to 5' exonuclease proofreading activity that corrects misincorporated bases during synthesis, producing blunt-ended PCR products with significantly lower error rates than standard Taq polymerase.
